Site-specific recombination of a transgene in fertilized eggs by transient expression of Cre recombinase.
An efficient method of transgene modulation in fertilized eggs has been developed that uses the Cre/loxP recombination system. Twelve transgenic mouse lines carrying a chicken beta-actin promoter-loxP-chloramphenicol acetyltransferase (CAT) gene-loxP-beta-galactosidase gene construct were produced. After selection of the line showing the highest expression of the CAT gene in a variety of tissue...
متن کاملMicroinjection of cre recombinase RNA induces site-specific recombination of a transgene in mouse oocytes.
We have developed a strategy for producing single copy transgenic mouse lines using Cre-loxP site specific recombination. The method is based on transient expression of the recombinase after injection of in vitro transcribed mRNA into the cytoplasm of fertilised eggs containing multiple copies of the transgene. The success rate of the recombination event is 100% (15 out of 15).

We report the use of the bacteriophage P1 Cre-lox system for generating conservative site-specific recombination between tobacco chromosomes. Two constructs, one containing a promoterless hygromycin-resistance gene preceded by a lox site (lox-hpt) and the other containing a cauliflower mosaic virus 35S promoter linked to a lox sequence and the cre coding region (35S-lox-cre), were introduced se...
متن کاملSite-specific DNA recombination in mammalian cells by the Cre recombinase of bacteriophage P1.
The Cre protein encoded by the coliphage P1 is a 38-kDa protein that efficiently promotes both intra- and intermolecular synapsis and recombination of DNA both in Escherichia coli and in vitro. Recombination occurs at a specific site, called lox, and does not require any other protein factors. The Cre protein is shown here also to be able to cause synapsis of DNA and site-specific recombination...
متن کاملGene deletion in urothelium by specific expression of Cre recombinase.
Urothelium that lines almost the entire urinary tract acts as a permeability barrier and is involved in the pathogenesis of major urinary diseases, including urothelial carcinoma, urinary tract infection, and interstitial cystitis. However, investigation of urothelial biology and diseases has been hampered by the lack of tissue-specific approaches. To address this deficiency, we sought to devel...
